White House press secretary Karoline Leavitt said on Thursday that Trump had discussed the condition to Saudi leaders on numerous previous occasions.
"He has said if they don't join the Abraham Accords, the deal is off," she said.
Trump's comments come come after criticism from experts that the US-Saudi deal announced on Wednesday could run the risk of future nuclear proliferation in an already unstable region.
According to the US Department of Energy, it includes a "peaceful nuclear co-operation agreement" and a "bilateral safeguards agreement".
"Together, these two agreements lay the legal foundation for a decades-long, multi-billion-dollar partnership that advances several priority economic and strategic objectives, including nuclear nonproliferation," a statement from the department said.
