Derbyshire Constabulary announced the road closure at around 11.30pm on Tuesday, August 11.
The A6 at Matlock is closed between Midland Hotel and Fishpond due to the crash.
Derbyshire Police are at the scene of the incident.
A spokesperson for Derbyshire Constabulary said: "The A6 at Matlock , between Midland Hotel and Fishpond, is currently closed due to a serious collision.
