Stevens-Johnson syndrome (SJS) and toxic epidermal necrolysis (TEN), the most severe form of drug hypersensitivity response with a 50% fatality rate, are often linked to antiepileptic medications (AEDs). Aromatic AEDs accounted for nearly all AED-associated cases (96%; 95% CI, 92-99), most often carbamazepine (39%; 95% CI, 29-50), phenytoin (19%; 95% CI, 11-28), and lamotrigine (15%; 95% CI, 8-22). The percentage of AED-associated SJS/TEN varied significantly by region, from 42% (95% CI, 26-59) in West Asia to 10% (95% CI, 0-33) in Africa. Carbamazepine, phenytoin, and lamotrigine were the most often implicated aromatic compounds in nearly all AED-associated cases.
