The FDA has granted Breakthrough Therapy designation to olomorasib as a monotherapy for adults with advanced pancreatic cancer who have already received at least one systemic therapy and whose tumors carry a KRAS G12C mutation. And the mutation involved is uncommon in pancreatic cancer, occurring in roughly 1 to 2 percent of cases, which means most people with pancreatic cancer would not be eligible even if the drug were approved tomorrow. Lilly notes that the five-year survival rate for metastatic pancreatic cancer remains below 5 percent. FDA accepted a new drug application in July 2026 for daraxonrasib, a RAS inhibitor from Revolution Medicines, in previously treated metastatic pancreatic cancer. KRAS G12C occurs in roughly 1 to 2 percent of pancreatic cancers, though KRAS mutations overall are found in about 90 percent.
