Avengers: Endgame held on the top spot as the number one all-time domestic opening with its $US357.1 million opening in April 2019.
Globally, the superhero movie took a worldwide opening amount of $US927 million ($A1.3 billion), becoming the second-best global launch.
The success of the film far surpassed the $US195 million and $US465 million global Marvel Studios had predicted.
Spider-man: Brand New Day's success means the Marvel Cinematic Universe has now surpassed the $US33.3 billion mark.
The film is Tom Holland's seventh within the MCU - his fourth solo outing as the web slinger.
