The attacker has linked up with old Vale boss Andy Crosby at League Two Tranmere Rovers.
Port Vale remain rooted to the bottom of the League One table as the January transfer window slams shut on Monday.
A Port Vale spokesman said: "We can confirm that James Plant has joined Sky Bet League Two side Tranmere Rovers on a permanent deal.
Everybody at Port Vale would like to wish James the very best for the future and thank him for his efforts during his time at Vale Park."
